CPK-EF refers to the EF-hand-containing regulatory region characteristic of plant calcium-dependent protein kinases (CPKs), which are cytosolic kinases that often associate with cellular membranes (frequently the plasma membrane) through N-terminal lipid modifications that help position the enzyme near upstream calcium signals and downstream targets. Structurally, a typical CPK is a single polypeptide composed of an N-terminal variable segment, a central serine/threonine protein kinase catalytic domain, an adjacent autoinhibitory/pseudosubstrate segment, and a C-terminal calmodulin-like domain that contains EF-hand motifs (the CPK-EF portion) that bind Ca2+. Ca2+ binding to these EF-hands triggers conformational changes that relieve autoinhibition and activate the kinase, enabling phosphorylation of substrate proteins. Functionally, CPKs act as Ca2+ sensors and signal transducers that couple stimulus-evoked Ca2+ transients to phosphorylation-driven changes in cellular activity, contributing broadly to regulation of plant growth and development and to responses to biotic and abiotic stresses by modulating signaling networks and gene expression programs.
