CPX1 (complexin 1) encodes a small, predominantly cytosolic neuronal protein enriched at synapses where it associates with presynaptic release sites and synaptic vesicle-proximal membranes through interactions with the assembled SNARE fusion machinery. The protein is largely helical and organized into an N-terminal region that supports evoked release, an accessory alpha-helix that can inhibit premature "zippering" of the SNARE bundle (fusion clamp activity), and a central SNARE-binding alpha-helix that docks along the SNARE complex; its C-terminal region contributes to efficient targeting/engagement near membranes and can tune inhibitory control of spontaneous fusion. Functionally, CPX1 is a key regulator of synaptic vesicle exocytosis, coupling to neuronal SNARE complexes (including syntaxin-1, SNAP25, and synaptobrevin/VAMP) to both promote fast Ca2+-triggered neurotransmitter release and suppress inappropriate spontaneous fusion by stabilizing specific prefusion SNARE states; in addition to presynaptic roles, complexin-dependent SNARE regulation has also been implicated in activity-regulated postsynaptic exocytosis events such as AMPA receptor insertion during synaptic plasticity.
