CURT1A encodes a small, nuclear-encoded chloroplast protein that is imported into chloroplasts and accumulates in the thylakoid membrane system, where it is highly enriched at grana margins (the strongly curved rim regions that connect stacked grana membranes to adjacent lamellae). CURT1A is an integral thylakoid membrane protein with an N-terminal chloroplast transit peptide, followed by an N-terminal amphipathic helix and two closely spaced transmembrane helices that support homo- and hetero-oligomerization with other CURT1 family members. Functionally, CURT1A is a key determinant of thylakoid ultrastructure: changing CURT1A abundance shifts grana architecture, and purified CURT1A can oligomerize and directly bend membranes in vitro (e.g., inducing liposome tubulation), supporting a primary role in generating/maintaining the extreme curvature characteristic of grana margins. By shaping grana curvature and thereby influencing the partitioning and connectivity of thylakoid subdomains, CURT1A contributes to proper organization of the photosynthetic membrane network and efficient operation and remodeling of the light reactions under changing conditions.
