CURT1D encodes a small intrinsic chloroplast thylakoid membrane protein that accumulates within the thylakoid system and is enriched at grana margins, the highly curved regions connecting appressed grana membranes to stromal lamellae. The protein is synthesized with an N-terminal chloroplast targeting peptide and is predicted to insert into thylakoids via two closely spaced transmembrane helices, with an additional N-terminal amphipathic helix proposed to couple membrane binding to curvature generation; CURT1 family members also form oligomers in vivo. Functionally, CURT1D contributes to shaping higher-order thylakoid architecture by promoting and stabilizing strong membrane curvature at grana margins, thereby influencing grana morphology and the overall 3-dimensional organization of the photosynthetic membrane network; across the CURT1/CurT family, this curvature activity is evolutionarily conserved and is linked to proper thylakoid system organization, with broader connections to thylakoid remodeling and partitioning during organelle division in oxygenic photosynthesizers.
