CXCL13 Antibody (17-7981-82) in Flow

BALB/c mouse resident peritoneal macrophages were stained with CD11b Monoclonal Antibody, FITC (Product # 11-0112-82) and F4/80 Monoclonal Antibody, eFluor 450 (Product # 48-4801-82). The cells were then fixed and permeabilized using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and protocol, and stained with 0.06 µg of Rat IgG2a kappa Isotype Control, APC (Product # 17-4321-81) (left) or 0.06 µg of CXCL13 Monoclonal Antibody, APC (right). CD11b positive cells were used for analysis.

Description: This DS8CX13 monoclonal antibody recognizes mouse CXCL13 also known as BCA-1 and Angie.

Applications Reported: This DS8CX13 antibody has been reported for use in flow cytometric analysis.

Applications Tested: This DS8CX13 antibody has been tested by flow cytometric analysis of resident peritoneal macrophages. This may be used at less than or equal to 0.125 µg per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.

CXCL13, also known as BCA-1 and Angie, is a small chemokine strongly expressed in the secondary lymphoid organs. It preferentially promotes the migration of B lymphocytes (both B-1 and B-2) to the follicles of the spleen, lymph nodes, and Peyer's patches. It is produced by macrophages, and in mouse the highest constitutive producers include large peritoneal macrophages, Kuppfer cells and lung macrophages (both CD11c+ and CD11c-). The cellular ligand for CXCL13 is CXCR5 also known as BLR-1 (Burkitt's Lymphoma Receptor 1). CXCR5 is expressed on B cells, Tfh cells and, to a lower degree, some memory T cell populations. CXCL13 plays an important role in the development of chronic lymphocytic leukemia (CCL). CXCL13 is also a plasma biomarker for germinal center activity and generation of broadly neutralizing antibodies against HIV.
