CesA8 encodes a plasma membrane-localized cellulose synthase catalytic subunit that functions in cellulose deposition during secondary cell wall formation, particularly in developing xylem, where loss of function causes characteristic irregular xylem with weakened, collapsed vessels. The CesA8 protein is a large, multi-pass membrane glycosyltransferase with an N-terminal cytosolic region containing a Cys-rich zinc-finger-like motif implicated in cellulose synthase complex assembly, followed by a central cytosolic catalytic domain typical of processive GT2 enzymes (including conserved motifs required for beta-1,4-glucan polymerization) and multiple C-terminal transmembrane helices that anchor the enzyme and support extrusion of the growing glucan chain into the cell wall. Biologically, CesA8 acts as one of the three nonredundant secondary-wall CesA isoforms that co-assemble into cellulose synthase complexes at the cell surface to synthesize cellulose microfibrils that provide mechanical strength and proper architecture to lignified secondary walls; consistent with its role in wall integrity, CesA8 disruption can also trigger compensatory stress-associated responses.
