ChlG encodes chlorophyll synthase, an integral membrane prenyltransferase that localizes to photosynthetic membranes (thylakoid membrane in oxygenic phototrophs and the cytoplasmic/intracytoplasmic membrane system in many bacteria), where it is positioned to couple pigment biosynthesis with assembly of chlorophyll-binding complexes. The protein belongs to the UbiA-type prenyltransferase superfamily and is a hydrophobic, multi-pass membrane enzyme whose catalytic core contains conserved Asp-rich motifs characteristic of this family that coordinate prenyl diphosphate chemistry. Functionally, ChlG catalyzes the terminal step of chlorophyll biosynthesis by esterifying chlorophyllide with a tetraprenyl alcohol moiety donated from phytyl diphosphate or geranylgeranyl diphosphate, producing (phytylated or geranylgeranylated) chlorophyll and releasing diphosphate. In cyanobacteria, ChlG is found in pigment-protein assemblies that include small high light-inducible proteins (e.g., HliD/HliC) and can associate with the YidC/Alb3 membrane insertase and the PSII biogenesis factor Ycf39, supporting a model in which ChlG not only synthesizes chlorophyll but also helps channel newly made chlorophyll toward cotranslational insertion and maturation of nascent photosystem apoproteins during acclimation and stress.
