Conglutin gamma encodes a lupin seed protein that accumulates predominantly in developing seed tissues and is stored in protein bodies/vacuoles as a secretory-pathway product synthesized with an N-terminal signal peptide typical of storage globulins. The mature gamma conglutin polypeptide is post-translationally processed into two subunits (commonly termed alpha and beta chains) that remain covalently linked by at least one disulfide bond, and the protein assembles into a quaternary oligomer consistent with the 7S basic globulin family; structural analyses show strong overall fold similarity to plant glycoside-hydrolase inhibitor proteins, although gamma conglutin does not exhibit the canonical inhibitory activity against representative fungal GH12 endo-glucanases. Biologically, gamma conglutin is considered part of the seed reserve proteome supporting early seedling growth, and its tightly regulated, seed-specific transcription supports this storage role; additionally, its inducible expression upon elicitation (e.g., chitosan treatment) and emerging biochemical data suggesting selective inhibition of a GH2 beta-mannosidase are consistent with a potential ancillary function in seed defense or modulation of cell-wall carbohydrate metabolism during germination.
