DCL2 encodes a predominantly nuclear, Dicer-like RNase III endoribonuclease that functions in RNA interference pathways by recognizing double-stranded RNA substrates and processing them into small interfering RNAs, with a prominent role in antiviral silencing that becomes especially evident when the related DCL4 pathway is compromised; it can also contribute to endogenous small-RNA production, including trans-acting siRNA biogenesis, and has been implicated in maintaining aspects of DNA methylation-linked silencing. The DCL2 protein is a large, multidomain enzyme with an N-terminal DExD/H-box helicase module (ATP-dependent RNA remodeling), followed by a DUF283 region, a PAZ domain that helps engage RNA termini, two tandem RNase III catalytic domains that form the dimeric active center for staggered dsRNA cleavage, and C-terminal double-stranded RNA-binding domain(s) that stabilize substrate interactions; multiple alternatively spliced transcript variants are reported, consistent with potential isoform-level modulation of activity and/or regulation.
