Detection of human DDI2 by western blot. Samples: Whole cell lysate (50 µg) from HeLa, HEK293T, and Jurkat cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-DDI2 antibody (Product # A304-629A) (lot A304-629A-1) used for WB at 0.1 µg/mL. Detection: Chemiluminescence with an exposure time of 3 minutes.

Aspartic protease that mediates the cleavage of NFE2L1/NRF1 at 'Leu-104', thereby promoting release of NFE2L1/NRF1 from the endoplasmic reticulum membrane. Ubiquitination of NFE2L1/NRF1 is a prerequisite for cleavage, suggesting that DDI2 specifically recognizes and binds ubiquitinated NFE2L1/NRF1.
