DGAT2A encodes an acyl-CoA:diacylglycerol acyltransferase-family enzyme that localizes predominantly to the endoplasmic reticulum (ER) and can redistribute to the surface of cytosolic lipid droplets during fatty-acid loading, consistent with a role at ER-lipid droplet interfaces that support neutral-lipid deposition. The protein is an integral membrane acyltransferase predicted to contain two transmembrane helices, with a short luminal loop connecting them, while the catalytically important regions reside in a larger membrane-associated cytosolic domain; DGAT2-family proteins can also assemble into higher-order complexes and physically associate with upstream acyltransferases that generate diacylglycerol, helping couple substrate supply to triacylglycerol production. Functionally, DGAT2A catalyzes the terminal, committed step of triacylglycerol synthesis by transferring a fatty acyl group from long-chain acyl-CoA to diacylglycerol, thereby promoting triacylglycerol accumulation and lipid droplet growth; this activity supports cellular energy storage and buffering of excess fatty acids, and is implicated in tissue lipid handling in metabolic contexts where neutral-lipid synthesis and droplet expansion are dynamically regulated.
