DHAR1 encodes a dehydroascorbate reductase that functions as a soluble, cytosolic enzyme in Arabidopsis thaliana, contributing to cellular redox homeostasis by supporting the ascorbate-glutathione cycle. The DHAR1 protein is a small (~213 aa) glutathione-dependent oxidoreductase with a thioredoxin-like fold and glutathione S-transferase-like N- and C-terminal regions typical of plant DHARs, consistent with its role in binding glutathione and reducing dehydroascorbate back to ascorbate. Genetically, loss of DHAR1 reduces total DHAR activity in leaves and compromises maintenance of the ascorbate redox state during photooxidative stress, leading to heightened stress symptoms compared with wild type, indicating DHAR1 is a key enzymatic component for ascorbate recycling and protection against reactive oxygen species in planta.
