DOT1-like histone lysine methyltransferase (DOT1L) is a gene located on chromosome 19q13.12, encoding an enzyme responsible for methylating lysine 79 on histone H3, a critical modification influencing chromatin structure and gene expression. Unlike most histone methyltransferases, DOT1L does not contain a SET domain and uniquely catalyzes H3K79 methylation, which plays pivotal roles in the regulation of transcription and maintenance of genomic stability. Expression of DOT1L is vital for various biological processes, including cell cycle progression, development, and differentiation, particularly in hematopoietic cells. Aberrations in DOT1L activity have been associated with mixed-lineage leukemia (MLL), where DOT1L-mediated H3K79 methylation contributes to leukemogenesis by influencing gene expression related to proliferation and survival. Due to its involvement in cancer progression, DOT1L is being investigated as a therapeutic target, with inhibitors developed to specifically modulate its enzymatic activity, offering potential for treating MLL and other malignancies.
