DRP5B encodes a plant-specific dynamin-related mechanochemical GTPase that is primarily cytosolic but is recruited to organelle surfaces, where it assembles into ring-like structures at constriction sites on the cytosolic face of the chloroplast outer envelope to drive membrane fission during plastid binary division; loss of DRP5B causes impaired constriction and enlarged, dumbbell-shaped chloroplasts, consistent with a direct role in generating constrictional force. Structurally, DRP5B is a dynamin-family protein with the canonical GTPase domain and additional dynamin-like assembly/effector regions that support self-oligomerization and nucleotide-dependent remodeling of the division ring, with GTP hydrolysis coupled to ring turnover and productive fission. Beyond chloroplasts, DRP5B also localizes to peroxisomes and contributes to peroxisome division and function, interacting with itself and with established peroxisome fission/elongation components (including FIS1 proteins, DRP3 proteins, and multiple PEX11 isoforms), supporting a model in which DRP5B acts cooperatively within a shared organelle division network to execute late-stage scission on chloroplast and peroxisome membranes.
