Store lyophilized/reconstituted at -20°C; once reconstituted make aliquots to avoid repeated freeze-thaw cycles. Please remember to spin the tubes briefly prior to opening them to avoid any losses that might occur from material adhering to the cap or sides of the tube.

Deg1 encodes a soluble tRNA pseudouridine(38/39) synthase that localizes to both the nucleus and cytoplasm and acts on cytoplasmic as well as mitochondrial tRNAs. The protein is a basic ~442 amino acid enzyme that belongs to the TruA/Pus3-like family of stand-alone pseudouridine synthases and contains a conserved catalytic core in which an essential aspartate residue mediates uridine isomerization. Functionally, Deg1 catalyzes formation of pseudouridine at positions 38 and 39 within the tRNA anticodon stem-loop, a conserved modification important for maintaining efficient and accurate translation; loss of Deg1 activity reduces pseudouridylation at these sites, compromises growth particularly at elevated temperature, and has been linked to altered translational recoding (e.g., frameshifting/readthrough) and downstream nuclear phenotypes consistent with translation-dependent effects on genome maintenance.
