Store lyophilized/reconstituted at -20°C; once reconstituted make aliquots to avoid repeated freeze-thaw cycles. Please remember to spin the tubes briefly prior to opening them to avoid any losses that might occur from material adhering to the cap or sides of the tube.
For reconstitution add 200 µL of sterile water.
Specific Species Reactivity: Arabidopsis thaliana, Chlamydomonas reinhardtii, Hordeum vulgare, Nicotiana tabacum, Solanum lycopersicum, Synechocystis sp. PCC 6803
Deg5 encodes a chloroplast-localized Deg/HtrA family ATP-independent serine endopeptidase that accumulates in the thylakoid lumen, where it is peripherally associated with the lumenal face of thylakoid membranes and acts in photosynthetic protein quality control. Structurally, Deg5 is distinguished among plant Deg proteases by comprising essentially a single trypsin-like protease domain (lacking a C-terminal PDZ regulatory domain) and assembling as an oligomer; crystallographic analysis of Arabidopsis Deg5 revealed a trimeric architecture with the active site formed by a His-Asp-Ser catalytic triad positioned between two beta-barrel subdomains, and the trimer contains bound calcium ions within a central channel, consistent with regulated lumenal proteolysis. Functionally, Deg5 collaborates with the lumenal Deg8 protease in the photosystem II repair cycle under photoinhibitory/high-light stress, contributing to endoproteolytic cleavage of damaged reaction-center components-most notably facilitating degradation of photodamaged D1 protein to enable its in situ replacement and restoration of photosystem II activity.
