Deg8 encodes a chloroplast HtrA-family, ATP-independent serine endopeptidase that is imported into the thylakoid lumen, where it associates peripherally with thylakoid membranes and functions in protein quality control during photosystem II (PSII) repair under excess light. The Deg8 polypeptide is organized as an N-terminal trypsin-like serine protease domain followed by a C-terminal PDZ domain that supports substrate recognition and/or assembly, and structural studies show Deg8 adopts an HtrA-like fold and oligomerizes (with Deg5) into higher-order complexes that position the catalytic triad for proteolysis. Biologically, Deg8 contributes to the PSII repair cycle by catalyzing endoproteolytic cleavage of photodamaged D1 (PsbA), generating characteristic N- and C-terminal fragments and thereby facilitating subsequent processive degradation and replacement of D1; loss of Deg8 (especially together with Deg5) slows D1 turnover and increases sensitivity to photoinhibition, consistent with a role in maintaining thylakoid proteostasis during high-light stress.
