EDS1 encodes an intracellular, predominantly nucleocytoplasmic protein that can accumulate in the nucleus during immune activation, consistent with a role in transcriptional defense reinforcement. The protein is composed of an N-terminal lipase-like alpha/beta-hydrolase fold (class 3 lipase-like domain) that resembles catalytic lipases but is generally described as non-catalytic in immune signaling contexts, coupled to a C-terminal region that is essential for activity and contributes to formation of higher-order signaling-competent complexes. Functionally, EDS1 is a central regulatory node in plant innate immunity, required for robust basal resistance to biotrophic and hemi-biotrophic pathogens and for effector-triggered immunity mediated by Toll/interleukin1 receptor-type NLR receptors. EDS1 acts through obligate, molecularly distinct heterodimers with PAD4 or SAG101 that occupy different intracellular pools and provide nonredundant signaling outputs, collectively promoting salicylic acid-associated defense programs, pathogen-restricting responses, and appropriate cell-death-associated immunity when triggered by intracellular immune receptors.
