FBA class 1 encodes a cytosolic fructose-1,6-bisphosphate (FBP) aldolase of central carbon metabolism that catalyzes the reversible cleavage of FBP into glyceraldehyde-3-phosphate and dihydroxyacetone phosphate, thereby supporting glycolysis and gluconeogenesis (and, in photosynthetic lineages, related carbon-assimilation reactions). In archaeal systems where this gene has been biochemically and structurally characterized, the encoded enzyme is a thermostable, metal-independent class I aldolase that proceeds via a Schiff-base intermediate, consistent with an essential catalytic lysine and a TIM-barrel-type fold; quaternary structure is oligomeric, with reported assemblies including homodecamers in solution for Thermococcus enzymes and decameric arrangements resolved in crystallographic studies of archaeal-type class I aldolases. Functionally, these properties position FBA class 1 as a key, high-flux metabolic enzyme that links hexose-phosphate utilization to triose-phosphate production and can be regulated by small metabolites that bias activity toward the catabolic direction under relevant physiological conditions.
