Ferredoxin 1 (FDX1) encodes a nuclear-encoded mitochondrial protein that is synthesized as a precursor with an N-terminal mitochondrial targeting sequence and is imported into the mitochondrial matrix, where it functions as a small soluble electron-transfer factor. Structurally, FDX1 is a ferredoxin-type iron-sulfur protein in the mitochondrial 2Fe-2S ferredoxin family, binding a 2Fe-2S cluster via conserved cysteine residues within a ferredoxin fold that supports reversible redox cycling and transient protein-protein interactions. Biologically, FDX1 shuttles electrons from the NADPH-dependent ferredoxin reductase (FDXR) to multiple mitochondrial type I cytochrome P450 enzymes, thereby supporting key monooxygenase reactions central to steroid hormone biosynthesis and related pathways (including steps in vitamin D and bile acid metabolism). Beyond steroidogenic P450 support, experimental evidence indicates FDX1 has distinct target specificity from the paralog FDX2 and can contribute electrons to additional mitochondrial biosynthetic processes, including heme a and lipoyl cofactor biosynthesis, consistent with a broader role in mitochondrial redox chemistry and respiratory competence.
