FLS2 encodes a plasma membrane-localized leucine-rich repeat receptor kinase that functions at the cell surface to detect bacterial flagellin-derived epitopes (notably the conserved peptide flg22) and initiate pattern-triggered immunity. The protein is a single-pass transmembrane receptor with a large extracellular ectodomain composed of multiple leucine-rich repeats (commonly described as 28 LRRs) that mediate ligand perception, flanked by conserved cysteine motifs important for proper maturation and receptor competence, and a cytosolic serine/threonine kinase domain required for downstream signaling. Upon flg22 perception, FLS2 rapidly forms an activated receptor complex with the co-receptor kinase BAK1, enabling transphosphorylation-dependent signal propagation that drives early immune outputs such as oxidative burst, defense gene induction, and broad antibacterial resistance. Loss-of-function alleles disrupt flagellin binding and/or signaling, demonstrating that both the extracellular LRR region and intracellular kinase activity are essential for full receptor function in vivo.
