FNRL encodes a plastid-localized, soluble oxidoreductase that resides predominantly in the chloroplast stroma and belongs to the plant-type ferredoxin-NADP+ oxidoreductase (FNR) superfamily. It is a nuclear-encoded protein predicted to be imported into chloroplasts and adopts the canonical two-domain flavoprotein architecture of this family, comprising an N-terminal FAD-binding domain and a C-terminal NADP+-binding alpha/beta domain, consistent with an FAD-dependent redox enzyme; biochemical and structural analyses indicate an NADPH-dependent activity and a cofactor-binding environment resembling bacterial-type ferredoxin-NADP+ reductases rather than the classical photosynthetic FNRs, including sequence features suggesting altered (or weak) interaction with the major photosynthetic ferredoxin. Functionally, genetic evidence shows that loss of FNRL alters stress-responsive signaling linked to mitochondria and confers tolerance to the cellulose biosynthesis inhibitor isoxaben, with phenotypes consistent with FNRL acting upstream of isoxaben-triggered cellular responses and intersecting with mitochondrial retrograde signaling pathways, implying that FNRL contributes to redox homeostasis and organelle-to-nucleus communication rather than serving as a primary photosynthetic electron-transfer enzyme.
