The antibody may be stored at -20°C for one year in its original formulation. Additionally, antibody may be stored at 2°C to 8°C for up to 1 month without detectable loss of activity. Avoid repeated freeze-thaw cycles of the diluted antibody.
For reconstitution add 50 µL of sterile water.
Specific Species Reactivity: Arabidopsis thaliana
In Arabidopsis thaliana, Fumarase 1 (FUM1) and Fumarase 2 (FUM2) encode class I fumarate hydratase enzymes that occupy distinct subcellular locations: FUM1 is targeted to mitochondria via an N-terminal targeting sequence, whereas FUM2 lacks mitochondrial targeting information and functions in the cytosol. Both proteins are soluble enzymes expected to assemble into multimeric complexes typical of fumarases and catalyze the reversible interconversion of fumarate and malate, thereby linking organic-acid pools with central carbon metabolism. Mitochondrial FUM1 supports the tricarboxylic acid cycle and is required for viability, consistent with an essential role in respiratory energy metabolism. In contrast, cytosolic FUM2 is dispensable under standard growth conditions but is critical for the large diurnal accumulation of fumarate in leaves, acting as a major sink for fixed carbon and helping coordinate carbon-nitrogen balance; loss of FUM2 reduces fumarate accumulation, shifts organic-acid partitioning toward malate, and compromises growth particularly under high nitrogen. Biochemical analyses indicate that FUM1 and FUM2 have broadly similar catalytic capacities yet differ in regulatory behavior, supporting a model in which compartmentation and differential regulation tailor fumarase activity to mitochondrial respiration versus cytosolic fumarate storage and acclimatory metabolism, including responses to temperature stress.
