FtsH1 and FtsH5 encode closely related, chloroplast-targeted FtsH family proteases that are integral to the thylakoid membrane, with their large soluble regions exposed on the stromal side. They are membrane-bound AAA+ ATP-dependent Zn2+ metalloproteases: an N-terminal membrane anchor (typically one or two transmembrane helices) is followed by a central AAA ATPase domain that powers substrate unfolding/translocation, and a C-terminal protease domain containing the conserved HEXXH metal-binding motif required for zinc-dependent proteolysis. In Arabidopsis, FtsH1 and FtsH5 comprise the "type A" subunits that assemble with "type B" partners (notably FtsH2/FtsH8) into a heterohexameric FtsH complex in the thylakoid, a major membrane quality-control protease system essential for chloroplast biogenesis and thylakoid maintenance. Functionally, the complex removes damaged and misfolded thylakoid proteins and is particularly important for the photosystem II repair cycle by promoting turnover of photodamaged reaction-center components (including D1), thereby sustaining photosynthetic performance under light stress; genetic evidence indicates substantial redundancy between the type A subunits, with loss of both FtsH1 and FtsH5 causing severe chloroplast developmental defects.
