FtsH2 and FtsH8 are nuclear-encoded, chloroplast-targeted, integral thylakoid membrane FtsH-family AAA+ metalloproteases that accumulate on the stromal side of the membrane as part of the major thylakoid FtsH protease complex. Each protein is predicted to contain an N-terminal chloroplast transit peptide followed by one (or two closely spaced) N-terminal transmembrane helix region(s) anchoring the enzyme in the thylakoid, with a large C-terminal stromal domain comprising an AAA ATPase module coupled to an M41 zinc metalloprotease domain bearing the characteristic HEXXH catalytic motif typical of active FtsH proteases. In Arabidopsis, FtsH2 and FtsH8 constitute the closely related "type B" subunits that assemble with "type A" subunits into a hetero-hexameric membrane complex; genetic analyses show FtsH2 and FtsH8 act redundantly and are jointly required for normal chloroplast biogenesis and thylakoid maintenance, with combined loss causing severe defects. Functionally, the thylakoid FtsH complex is a central ATP-dependent quality-control protease that recognizes, unfolds, and processively degrades damaged or misassembled thylakoid proteins, with a key established role in the photosystem II repair cycle by promoting turnover of photodamaged D1, thereby supporting recovery from photoinhibition and sustaining photosynthetic performance.
