GLDP encodes the mitochondrial P-protein of the glycine cleavage system, a soluble matrix enzyme that is imported via an N-terminal mitochondrial targeting sequence and functions as a pyridoxal 5-prime-phosphate (PLP)-dependent aminotransferase-like decarboxylase. The mature protein is a large PLP-binding enzyme with conserved catalytic motifs typical of class I/II aminotransferase fold enzymes and forms a functional homodimer, creating an active-site architecture that binds glycine and coordinates PLP to support catalysis; structural work on P-proteins supports a dimeric organization and highlights conserved residues near the active site that regulate access to the PLP/glycine-binding pocket. Biologically, the P-protein catalyzes the initial step of glycine degradation in one-carbon metabolism by decarboxylating glycine and transferring the resulting aminomethyl moiety to the lipoyl domain of the H-protein, thereby coupling glycine catabolism to folate-dependent one-carbon units and cellular redox/energy metabolism; loss of P-protein activity disrupts glycine clearance and impairs flux through mitochondrial one-carbon pathways.
