GSDMD Antibody (PA5-115330) in ICC/IF
Immunocytochemistry analysis of GSDMD in HeLa cells (LPS treated 4h). Samples were treated with PFA, permeabilized in 0.1% Triton X-100, blocked in 10% serum (45 min at 25°C), and incubated with polyclonal antibody (Product # PA5-115330) at a dilution of 1:200 (1 hr at 37°C). Secondary staining was applied with mouse anti-beta tubulin; AlexaFluor 594 conjugated goat anti-rabbit IgG (Red); AlexaFluor 88 conjugated goat anti-mouse IgG (Green) and DAPI (blue) using a dilution of 1:200 (1 hr at 37°C).

Antibody detects endogenous levels of total GSDMD protein, including precursor and the cleaved N-terminal fragment.
靶标信息
GSDMD (gasdermin-D) promotes pyroptosis in response to microbial infection and danger signals. It is produced by the cleavage of gasdermin-D by inflammatory caspases CASP1 or CASP4 in response to canonical, as well as, non-canonical (such as cytosolic LPS) inflammasome activators. After cleavage, GSDMD moves to the plasma membrane where it strongly binds to inner leaflet lipids including phosphatidylinositol 4-phosphate, phosphatidylinositol (4,5)-biphosphate, phosphatidylinositol (3,4,5)-biphosphate, phosphatidic acid, and phosphatidylserine. GSDMD also exhibits bactericidal activity.
