GUN1 encodes a nucleus-encoded, chloroplast-localized regulatory protein that accumulates predominantly during early chloroplast biogenesis and associates with plastid gene-expression and proteostasis machineries. The protein is a member of the PPR-SMR class, comprising multiple pentatricopeptide repeat (PPR) motifs that support macromolecular interactions in plastids and a C-terminal small MutS-related (SMR) domain, a module linked in other contexts to nuclease-related activities. Functionally, GUN1 is a key integrator of plastid-to-nucleus retrograde signaling triggered by disrupted chloroplast development and impaired plastid gene expression; genetic and biochemical evidence indicates it coordinates nuclear photosynthesis-associated gene expression with chloroplast status by influencing plastid RNA metabolism (including transcriptional responses and RNA editing under signaling conditions) and by interacting with components involved in chloroplast protein homeostasis.
