Glibenclamide, also known as glyburide, is a second-generation sulfonylurea used primarily in the management of type 2 diabetes mellitus. It functions by stimulating insulin secretion through the inhibition of ATP-sensitive potassium channels on pancreatic beta cells, leading to increased intracellular potassium and calcium ion concentrations. The gene encoding for the ATP-sensitive potassium channel is located on chromosome 11, and its structure is crucial for the drug's mechanism of action. Glibenclamide is often combined with metformin hydrochloride to enhance glycemic control by addressing complementary mechanisms. Various formulations of glibenclamide exist, including modified-release versions, which aim to improve bioavailability and control drug release. These formulations are evaluated for bioequivalence and pharmacodynamics to ensure consistent therapeutic effects.
