A , B , Changes in AMPA receptor subunit expression of the AMPA receptor subunits GluR-A, GluR-B, GluR-C, and GluR-D in hippocampus ( A ) and cerebellum ( B ) of wild-type (WT) and GluR-C knock-out (GluR-C -/-) mice from P2 to P90. Total proteins of the hippocampus and the cerebellum were isolated, and for each sample, 10 mug of protein was separated on a 7.5% SDS-PAGE and transferred on nitrocellulose. The glutamate receptor subunits were visualized by selective antibodies and monitored by autoradiography. The alpha-subunit of the Ca 2+ /calmodulin-dependent protein kinase II (alpha-CaMKII) and betaIII-tubulin (betaIII-Tub.) was used as a positive control for developmental controlled gene expression in hippocampus and cerebellum, respectively. Loading was controlled by beta-actin. The experiment was repeated with at least three sets of mice. One representative example for each subunit is given. {KO}

Glutamate receptors are the predominant excitatory neurotransmitter receptors in the mammalian brain and are activated in a variety of normal neurophysiologic processes. These receptors are heteromeric protein complexes composed of multiple subunits, arranged to form ligand-gated ion channels. The classification of glutamate receptors is based on their activation by different pharmacologic agonists. The subunit encoded by this gene belongs to a family of AMPA (alpha-amino-3-hydroxy-5-methyl-4-isoxazole propionate)-sensitive glutamate receptors, and is subject to RNA editing (AGA->GGA; R->G). Alternative splicing at this locus results in different isoforms, which may vary in their signal transduction properties.
