Store lyophilized/reconstituted at -20°C; once reconstituted make aliquots to avoid repeated freeze-thaw cycles. Please remember to spin the tubes briefly prior to opening them to avoid any losses that might occur from material adhering to the cap or sides of the tube.

GLRX5 encodes a conserved mitochondrial glutaredoxin that is synthesized as a precursor bearing an N-terminal mitochondrial targeting sequence and is imported to the mitochondrial matrix, where it functions in iron-sulfur (Fe-S) cluster biogenesis and cellular iron homeostasis. The protein is a small monothiol glutaredoxin family member with a single glutaredoxin-like domain and a characteristic CGFS active-site motif that supports coordination/handling of a [2Fe-2S] cluster, consistent with a role as an Fe-S cluster transfer/assembly factor within the mitochondrial ISC machinery. Functionally, GLRX5 is required for maturation of mitochondrial Fe-S proteins and for maintaining proper distribution of iron between mitochondrial and cytosolic pools; loss of GLRX5 activity impairs Fe-S cluster formation, perturbs iron regulation, and in erythroid cells can secondarily compromise heme biosynthesis by disrupting Fe-S-dependent steps and iron-responsive regulatory pathways, causing autosomal recessive pyridoxine-refractory sideroblastic anemia.
