HDAC2 Antibody in Western Blot (WB) — Western blot analysis of HDAC2 on different lysates. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in PBS for 1 hour at room temperature. HDAC2 Recombinant Monoclonal Antibody (Product # MA5-42453) at 1:500 was used in 5% BSA at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:200,000 dilution was used for 1 hour at room temperature. Positive control: Lane 1: Hela cell lysate. Lane 2: K562 cell lysate. Predicted band size: 55 kDa. Observed band size: 60 kDa.

HDAC2 Antibody in Western Blot (WB) — Knockout of HDAC2 was achieved by CRISPR-Cas9 genome editing using LentiArray™ Lentiviral sgRNA (Product # A32042, Assay ID CRISPR1097024_LV) and LentiArray Cas9 Lentivirus (Product # A32064). Western blot analysis of HDAC2 was performed by loading 30 µg of HCT 116 wild type (Lane 1), HCT 116 Cas9 (Lane 2) and HCT 116 HDAC2 KO (Lane 3) whole cell extracts. The samples were electrophoresed using NuPAGE™ Novex™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with HDAC2 Recombinant Rabbit Monoclonal Antibody (SD0816) (Product # MA5-42453, 1:500 dilution) and Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:10,000 dilution) using the iBright™ FL1500 (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076). Loss of signal upon CRISPR mediated knockout (KO) using the LentiArray™ CRISPR product line confirms that antibody is specific to HDAC2.

HDAC2 Antibody in Western Blot (WB) — All lanes: Western blot analysis of HDAC2 with HDAC2 Recombinant Monoclonal Antibody (Product # MA5-42453) at 1:500 dilution. Lane 1: Wild-type Hela whole cell lysate (10 µg). Lane 2/3: HDAC2 knockout Hela whole cell lysate (10 µg). HDAC2 Recombinant Monoclonal Antibody was shown to specifically react with HDAC2 in wild-type Hela cells. No bands were observed when HDAC2 knockout sample were tested. Wild-type and HDAC2 knockout samples were subjected to SDS-PAGE. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM in TBST for 1 hour at room temperature. The primary antibody (1:500) was used in 5% BSA at room temperature for 2 hours. Goat Anti-Rabbit IgG-HRP Secondary Antibody at 1:300,000 dilution was used for 1 hour at room temperature.

HDAC2 belongs to the histone deacetylase family. Histone deacetylases act via the formation of large multiprotein complexes and are responsible for the deacetylation of lysine residues at the N-terminal regions of core histones (H2A, H2B, H3 and H4). This protein forms transcriptional repressor complexes by associating with many different proteins, including YY1, a mammalian zinc-finger transcription factor. Thus, it plays an important role in transcriptional regulation, cell cycle progression and developmental events. Alternative splicing results in multiple transcript variants.
