Nuclei were isolated from Arabidopsis thaliana leaves and resuspended with SDS loading buffer and then denatured at 95 o C for 10 min. HDT3 is a plant histone deacetylase of the HD2 family that modulates chromatin structure to regulate gene expression during development and stress responses. HDT3 removes acetyl groups from histone tails, leading to tighter nucleosome packing and transcriptional repression at specific loci, and it can also influence nucleolar organization and ribosomal RNA processing. Its expression and activity respond to hormones, abiotic stresses such as drought and salinity, and developmental cues, contributing to control of root and shoot growth, flowering, and stress acclimation. HDT3 often acts in complexes with other chromatin regulators and transcription factors, providing locus specific targeting, and genetic perturbation alters growth, phase transitions, and tolerance by reshaping transcriptional networks.
