HSP23.6 encodes a nuclear-encoded small heat shock protein that is post-translationally targeted to mitochondria, consistent with an N-terminal, mitochondria-targeting presequence and a mature protein of ~23.6 kDa. The gene model comprises two exons and produces a small heat shock protein of ~210 amino acids that contains the conserved Hsp20/alpha-crystallin domain characteristic of ATP-independent sHSP chaperones, typically flanked by more variable N- and C-terminal regions implicated in oligomerization and client binding. Biologically, HSP23.6 is stress responsive (notably induced by heat and other abiotic challenges) and is implicated in maintaining mitochondrial proteostasis during stress by acting as a holdase that stabilizes unfolding or aggregation-prone proteins; mitochondrial sHSP activity has been linked to protection of respiratory chain function under heat stress, and genetic perturbation of the mitochondrial sHSP set in Arabidopsis supports roles in normal growth and metabolism even in the absence of exogenous stress.
