HSP26.5 encodes a nuclear-encoded small heat shock protein of the HSP20/alpha-crystallin superfamily that is targeted to mitochondria, where it is strongly induced by elevated temperature and other stress conditions and contributes to organellar proteostasis. The protein is expected to contain an N-terminal mitochondrial targeting presequence followed by the conserved alpha-crystallin domain typical of small heat shock proteins, with variable N- and C-terminal regions that support oligomerization and substrate interactions. Functionally, HSP26.5 acts as an ATP-independent molecular chaperone that helps prevent irreversible aggregation of stress-denatured proteins in mitochondria and supports cellular homeostasis; genetic depletion studies in Arabidopsis indicate mitochondrial small heat shock proteins, including HSP26.5, have partially redundant roles that are important for normal growth and development even without externally applied stress.
