HTA9 encodes a non-canonical histone H2A variant of the H2A.Z class that localizes to the nucleus and is incorporated into chromatin as part of nucleosomes, thereby influencing local chromatin architecture and DNA accessibility. The gene is a compact, intron-containing locus (two exons) that produces an H2A-family protein expected to adopt the conserved histone fold required for H2A-H2B dimerization within the nucleosome core, while retaining H2A.Z-specific sequence features that distinguish it from canonical replication-coupled H2A. Biologically, HTA9 contributes to H2A.Z-dependent regulation of transcriptional programs that respond to developmental and environmental cues: genetic loss of H2A.Z function (including HTA9 in combination with other H2A.Z-encoding genes) leads to widespread gene misregulation, and has been linked to altered flowering-related regulation and temperature-responsive gene expression as well as changes in epigenetic states such as reduced transposon DNA methylation without broadly reducing genic methylation, consistent with a role for H2A.Z-containing nucleosomes in shaping plant chromatin function, genome stability, and signal-responsive gene control.
