HY5 encodes a nuclear basic leucine zipper (bZIP) transcription factor that functions as a central integrator of environmental light cues with developmental and physiological programs in Arabidopsis. The gene produces protein-coding transcripts whose product contains a conserved C-terminal bZIP DNA-binding/dimerization module (basic region plus leucine zipper) that mediates sequence-specific binding to ACGT-core light-responsive cis elements, particularly G-box motifs, thereby regulating transcription of downstream target promoters in response to illumination. Structural/biophysical work indicates HY5 is largely intrinsically disordered across its N-terminus, with conformational flexibility extending into the basic region, consistent with a regulatory protein that engages multiple partners and promoters. Functionally, HY5 acts predominantly as a positive regulator of photomorphogenesis, promoting light-dependent developmental transitions such as inhibition of hypocotyl elongation, seedling greening, and chloroplast development, and it contributes to stimulus-regulated root growth and patterning. HY5 abundance and activity are controlled post-translationally by light-dependent interaction with the E3 ubiquitin ligase COP1, which targets HY5 for proteasome-mediated degradation in darkness, enabling rapid tuning of light-responsive gene expression programs.
