Halofuginone is a derivative of febrifugine, an alkaloid originally isolated from the plant Dichroa febrifuga. It is used in veterinary medicine to prevent and treat protozoan diseases such as coccidiosis in poultry and cryptosporidiosis in calves. Halofuginone acts as a potent inhibitor of collagen alpha 1 (I) and matrix metalloproteinase 2 (MMP-2) gene expression, suppressing extracellular matrix deposition and cell proliferation. Its therapeutic applications extend to treating malaria, cancer, fibrosis, and inflammatory diseases, owing to its ability to inhibit prolyl-tRNA synthetase, a molecular target involved in these conditions.
