HydA encodes the catalytic subunit of an [FeFe]-hydrogenase that is typically a soluble enzyme found in the cytoplasm of anaerobic microorganisms, where it interfaces with cellular redox carriers to support fermentative and other anaerobic metabolic pathways. The HydA protein scaffold binds the defining H-cluster active site, which consists of a canonical [4Fe-4S] cubane linked through a cysteine thiolate to a distinctive 2Fe subcluster bearing non-protein ligands (including CO, CN, and a bridging dithiolate), forming the organometallic center responsible for catalysis; structural studies of apo-forms lacking the 2Fe module show a preformed [4Fe-4S] subcluster and an open pocket that accommodates stepwise insertion of the remaining 2Fe unit during maturation. Functionally, HydA catalyzes the reversible reduction of protons to molecular hydrogen (H2) and the oxidation of H2 back to protons and electrons, thereby helping balance intracellular redox and enabling hydrogen production or consumption depending on metabolic context; full activity depends on proper H-cluster assembly, in which general Fe-S biogenesis machinery provides the cubane and dedicated maturation factors assemble and deliver the 2Fe subcluster.
