Hygromycin B is an aminoglycoside antibiotic produced by the bacterium Streptomyces hygroscopicus. It functions by inhibiting protein synthesis, affecting both prokaryotic and eukaryotic cells. Hygromycin B binds to 16S rRNA, specifically near the A site at the top of helix 44, causing miscoding during translation, although less so than other aminoglycosides. It is particularly effective in halting yeast cell growth by preventing protein synthesis in cytoplasmic ribosomes. This antibiotic is used in molecular biology as a selection marker for genetically modified organisms due to its ability to kill bacteria, fungi, and higher eukaryotic cells. Structurally, hygromycin B is characterized by its interaction with the ribosomal RNA, differing from other antibiotics like paromomycin and streptomycin.
