IRT1 encodes an iron-regulated, integral membrane metal uptake transporter that is strongly induced in the outer cell layers of Arabidopsis roots under iron limitation and functions at the plant-soil interface. The IRT1 protein belongs to the ZIP family of metal transporters and is a multi-pass plasma-membrane protein with a conserved ZIP domain; it contains a prominent cytosolic loop implicated in metal sensing/turnover, including a histidine-rich motif and lysine residues that are targets for ubiquitin-mediated regulation. In root epidermal cells (including root hairs), IRT1 dynamically cycles between the plasma membrane and the trans-Golgi network/early endosomes, where constitutive endocytosis and monoubiquitin-dependent trafficking help prevent toxic metal overaccumulation while maintaining iron acquisition. Functionally, IRT1 is the major high-affinity entry route for ferrous iron (Fe2+) from the rhizosphere during iron starvation and can also transport other divalent metals such as manganese, zinc, cobalt, and cadmium, making its abundance and subcellular localization critical determinants of whole-plant metal homeostasis.
