Immunofluorescence analysis of Insulin gene enhancer protein ISL-1 was performed using 70% confluent log phase SH-SY5Y cells. The cells were fixed with 4% paraformaldehyde for 15 minutes, permeabilized with 0.1% Triton™ X-100 for 10 minutes, and blocked with 2% BSA for 45 minutes at room temperature. ISL1 Antibody in Immunocytochemistry (ICC/IF) — Immunofluorescence analysis of Insulin gene enhancer protein ISL-1 was performed using 70% confluent log phase SH-SY5Y cells. The cells were labeled with ISL1 Recombinant Rabbit Monoclonal Antibody (ARC0511) (Product # MA5-35062) at 1:200 dilution in 0.1% BSA, incubated at 4 degree celsius overnight and then labeled with Donkey anti-Rabbit IgG (H+L) Highly Cross-Adsorbed Secondary Antibody, Alexa Fluor™ Plus 488 (Product # A32790), (1:3,000), for 45 minutes at room temperature (Panel a: Green). Nuclei (Panel b: Blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: Red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing nuclear localization. Panel f represents merged image showing no signal in HeLa cells. Panel f represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

ISL1 Antibody in Immunohistochemistry (Paraffin) (IHC (P)) — Immunohistochemistry analysis of ISL1 in paraffin-embedded mouse fetal heart. Samples were incubated with ISL1 Monoclonal antibody (Product # MA5-35062) using a dilution of 1:100 (40x lens). Perform high pressure antigen retrieval with 10 mM citrate buffer pH 6.0 before commencing with IHC staining protocol.

ISL1 Antibody in Western Blot (WB) — Western blot was performed using ISL1 Recombinant Rabbit Monoclonal Antibody (ARC0511) (Product # MA5-35062) and a 42 kDa band corresponding to Insulin gene enhancer protein ISL-1 was observed across the panel tested except for SK-OV-3 which is reported to be low expressor of ISL1 and THP-1 and HeLa which are null expressors of ISL1. Whole cell extracts (30 µg lysate) of SH-SY5Y (Lane 1), IMR-32 (Lane 2), SK-O-V3 (Lane 3), THP-1 (Lane 4), HeLa (Lane 5) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX), 10 well. Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1,000) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:20,000) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Pico PLUS Chemiluminescent Substrate (Product # 34580).

ISL1 Antibody in Western Blot (WB) — Western blot analysis of ISL1 in extracts of various cell lines. Samples were incubated with ISL1 Monoclonal antibody (Product # MA5-35062) using a dilution of 1:1,000, followed by HRP Goat Anti-Rabbit IgG (H+L) at a dilution of 1:10,000. Lysates/proteins: 25 µg per lane. Blocking buffer: 3% nonfat dry milk in TBST. Detection: ECL Basic Kit. Exposure time: 30s.

ISL1 Antibody — Antibody specificity was demonstrated by detection of differential basal expression of the target across cell lines tested owing to their inherent genetic constitution. Relative expression of Insulin gene enhancer protein ISL-1 was observed in all cell lines except in THP-1 and HeLa using ISL1 Recombinant Rabbit Monoclonal Antibody (ARC0511) (Product # MA5-35062) in Western Blot. {RE}

This gene encodes a member of the LIM/homeodomain family of transcription factors. The encoded protein binds to the enhancer region of the insulin gene, among others, and may play an important role in regulating insulin gene expression. The encoded protein is central to the development of pancreatic cell lineages and may also be required for motor neuron generation. Mutations in this gene have been associated with maturity-onset diabetes of the young.
