KatG encodes a heme-dependent catalase-peroxidase that is primarily a soluble, intracellular enzyme, functioning in the bacterial cytosol where it detoxifies reactive oxygen species and interfaces with redox-active substrates. The protein is a large monomeric hemoprotein (approximately 80 kDa) belonging to the catalase-peroxidase family of peroxidase-like enzymes, built around a peroxidase-type fold that houses a b-type heme cofactor and supports access channels to the heme edge for small-molecule substrates; structural studies also support multiple, relatively weak surface-proximal binding sites for isoniazid near heme access regions. Functionally, KatG exhibits dual enzymatic activities: (i) catalase activity that disproportionates hydrogen peroxide to water and oxygen, helping protect the cell from oxidative stress encountered during host infection, and (ii) peroxidase activity that enables one-electron oxidation reactions and is critical for activation of the tuberculosis prodrug isoniazid, with katG variation being a major genetic basis of clinical isoniazid resistance. This catalytic versatility is linked to cooperation between the heme and a distinctive protein-derived Met-Tyr-Trp covalent adduct cofactor that supports efficient catalatic turnover while maintaining peroxidase-like chemistry.
