LEA6-1 encodes a late embryogenesis abundant (LEA) group 6 protein that accumulates predominantly in seeds and is inducible by water-deficit-related cues; the protein is primarily localized to the cytosol and can also be detected in the nucleus, consistent with the predominantly cytosolic/nucleocytosolic distribution reported for LEA6-family members in Arabidopsis. Structurally, LEA6-1 is a small, highly hydrophilic protein dominated by an intrinsically disordered conformational state in aqueous conditions, with the capacity to gain limited ordered secondary structure (notably helical propensity) under dehydration-mimicking environments; such disorder-to-order behavior is characteristic of LEA proteins and is thought to support reversible interactions with macromolecules and membranes during stress. Functionally, LEA6-1 is implicated in cellular protection during desiccation/osmotic stress by contributing to maintenance of the intracellular "glassy" state and preservation of protein and membrane integrity, thereby supporting seed longevity and enhancing tolerance of early germination/seedling growth to high osmolarity and salinity stress; collectively, these data support a role for LEA6-1 as a stress-responsive, intracellular stabilizer that helps plants withstand low-water availability and related abiotic stresses.
