LHY encodes a MYB-related DNA-binding transcription factor that localizes predominantly to the nucleus, consistent with its role in transcriptional regulation within the plant circadian system. The LHY protein is a single polypeptide containing an N-terminal MYB/SHAQKYF-class DNA-binding domain typical of plant single-repeat MYB factors, enabling sequence-specific promoter binding, and it can participate in higher-order complexes including homo- and heterodimerization with the closely related clock component CCA1. Functionally, LHY is a core element of the Arabidopsis circadian oscillator: its transcript abundance cycles with a morning phase, and perturbation of LHY dosage disrupts circadian rhythms and downstream photoperiodic outputs. Mechanistically, LHY contributes to interlocked transcriptional feedback loops by directly repressing sets of evening-phased clock and output genes via binding to clock-associated cis-elements, thereby shaping daily rhythms in physiology and development (including photoperiodic flowering timing), and genome-wide binding studies further support broader clock-to-physiology coupling such as circadian control of abscisic acid biosynthesis and signaling under stress conditions.
