Lnx1 (Ligand of Numb Protein X 1) protein is encoded by the Lnx1 gene located on chromosome 4 in humans and is known as PDZRN2 (PDZ domain-containing RING finger 2). It is through its RING finger and PDZ domains that Lnx1 forms oligomers and interacts with some of its target proteins such as JAM4 to form multiprotein complexes. Lnx1 is best studied as a E3 ubiquitin ligase that also contains the NPXY motifs which allows binding to the Phosphotyrosine binding domain (PTB) on Numb protein. This interaction has been shown to be responsible for ubiquitination of Numb, which prevents the proteasomal degradation of Notch intracellular domain (NICD) and thus positively regulates the Notch signaling pathway.
