LCA5 encodes lebercilin, an intracellular cilia/centrosome-associated protein that localizes prominently to the photoreceptor connecting cilium as well as to microtubules and centrioles/centrosome-related structures in cultured cells, consistent with a role at the interface of the ciliary axoneme and trafficking machinery. The gene produces alternatively spliced transcripts, and the encoded protein contains a characteristic lebercilin domain (as defined by conserved domain resources) within a largely non-enzymatic polypeptide that is thought to function through protein-protein interactions rather than catalytic activity. Functionally, lebercilin participates in ciliary processes required for retinal photoreceptor maintenance; affinity proteomics and cellular studies support that it links to the intraflagellar transport (IFT) machinery and that disease-associated mutations disrupt these interactions, leading to impaired IFT-dependent transport within photoreceptor cilia and consequent early-onset retinal degeneration.
