LHCB5 encodes a nuclear-encoded chloroplast protein that is imported into the chloroplast and becomes an integral component of the thylakoid membrane within photosystem II (PSII) antenna assemblies. The mature polypeptide belongs to the light-harvesting chlorophyll a/b-binding (LHC) family and is a multi-pass membrane protein with the characteristic chlorophyll a/b-binding fold of LHC antenna subunits, enabling stable association with chlorophylls and carotenoids in the lipid bilayer. Functionally, LHCB5 is a minor PSII antenna that contributes to efficient light capture and excitation-energy transfer to the PSII reaction center, and it also participates in photoprotective regulation, including nonphotochemical quenching. Structural and biochemical analyses of plants lacking LHCB5 indicate that it occupies a specific position that helps couple the PSII core with peripheral LHCII trimers in higher-order PSII supercomplexes (e.g., C2S2M2) and supports proper PSII macro-organization in the thylakoid membrane, with loss of LHCB5 altering supercomplex/megacomplex stability and organization without necessarily abolishing overall photosynthetic competence.
