Lysine tRNA ligase, also called Lysyl tRNA synthetase, is an essential enzyme that charges tRNA Lys with lysine to enable accurate translation of lysine codons during protein synthesis. It recognizes lysine and tRNA Lys, activates lysine with ATP to form lysyl AMP, and transfers the amino acid to the tRNA, ensuring fidelity via editing and specific identity elements on the tRNA. In plants, lysine tRNA ligase localizes to the cytosol and organelles via distinct isoforms, supporting both cytosolic and organellar translation, and its activity influences growth, stress responses, and development by sustaining protein synthesis capacity. Regulation occurs through expression changes, posttranslational modifications, and interactions with translation factors, and perturbation affects translational efficiency, amino acid homeostasis, and resilience under environmental stress.
