Specific Species Reactivity: Arabidopsis thaliana
Metacaspase-4 (MC4) encodes a cytosolic, Ca2+-responsive cysteine endopeptidase best characterized in Arabidopsis thaliana, where it acts as a damage-activated protease linking loss of plasma membrane integrity to rapid processing of intracellular immune peptides. MC4 is synthesized as an inactive zymogen whose core architecture comprises a caspase-like peptidase C14 domain and an extended internal linker region that enforces autoinhibition by occluding substrate access and positioning a linker lysine residue between catalytic residues; elevated Ca2+ triggers conformational switching and multiple autoprocessing events within the linker to generate an active enzyme. Biochemically, Arabidopsis type II metacaspases including MC4 cleave substrates after basic residues (arginine/lysine), rather than after aspartate as in animal caspases, consistent with a distinct substrate specificity in plants. Functionally, MC4 is necessary and sufficient for maturation of the wound signal Pep1 from its cytosolic precursor PROPEP1 following damage-induced Ca2+ influx, thereby promoting local defense signaling; more broadly, MC4 has been implicated as a stress-responsive protease contributing to regulated cell death and immune activation pathways in planta.
